I know offensively, that there must be 7 players on the LOS, and 5 of them must wear between #50 and #79.

But are there rules for defensive formations? I mean, could someone just line up with 1 nose tackle, 6 "stand up" linebackers, and 4 DBs? Or maybe the opposite? 6 defensive linemen, 1 linebacker, and 4 dbs.

What types of restrictive rules are there defensively?

As long as they are all on their own side of the line of scrimmage, they can be wherever they want.
